If you install CS6 and delete CS5 as part of the install, the NIK (and most other) plugins will transfer properly to CS6. The big problem comes from installing CS6 along side CS5, I think.
I have Lion on both my macs and all the NIK products are supported. I doubt that any of the complex plugins are yet set up for Mountain Lion.

I'm not running Mountain Lion on my Mac yet. What I did on my PC when installing 5 & 6 side by side was to launch the NIK installer and reinstall Color Efex Pro. When I did that, it recognized 6 and installed the plugin for that.
